At home, I was able to get TM running over my network quite easily using the old "TMShowUnsupportedNetworkVolumes 1" command. There wasn't even an issue about it, it just worked.
But here at work, I've got two iMacs that I'd like to do the same thing with, only over wired instead of wireless, and it's not working. Both machines can see the volume I'm pointing them to, but they won't sync to it. They go through the motions as if they were about to, but after "Making backup disk available" for a few seconds, they say that "The backup disk is not available."
I can't figure out why. Both machines have read/write access to the volume, it's format is Mac OS Extended (Journaled), and it's got tons 'o space. The only thing I can think of is that the machine the disk is attached to is running 10.4, but that shouldn't matter, eh?

That's definitely strange that TM is having trouble. It works fine here at home with two machines backing up to a drive that is attached to my Mini. My wife's does it over the wireless network even though the drive is not mounted on her machine. TM simply "finds" it. Maybe it is a Leopard/Tiger argument.
